John Fletcher Darby (December 10, 1803 - May 11, 1882) was a U.S. Representative from Missouri and 4th mayor of St. Louis, Missouri.

Born in Person County, North Carolina, Darby attended the public schools.He moved with his father to Missouri in 1818, where he worked on a farm.He moved to Frankfort, Kentucky, in 1825.He studied law.He was admitted to the bar and afterward practiced in St. Louis, Missouri.He served twice as mayor of St. Louis from 1835-1837 and 1840-1841.He served as member of the Missouri Senate in 1838.

Darby was elected as a Whig to the Thirty-second Congress (March 4, 1851-March 3, 1853).He returned to St. Louis and engaged in banking.He died near Pendleton, Missouri, May 11, 1882.He was interred in Calvary Cemetery, St. Louis, Missouri.
