In June 1913, at age 22, Pepper made it to the Major Leagues with the Detroit Tigers. In 1913, the Tigers were looking for a new third baseman to replace George Moriarty. Peploski played in two games for the Tigers, on June 24, 1913, and June 26, 1913. He had 2 singles and scored a run in 4 at bats, for a career batting average of .500. In two games at third base, Peploski had 1 assist and made no errors for a perfect 1.000 career fielding percentage. Despite good numbers in the two games he played, the Tigers chose Ossie Vitt as their third baseman of the future. Peploski's younger brother, Henry Peploski played third base for the Boston Braves in 1929. Henry, who was nicknamed "Pep" Peploski, played in six Major League games, 4 more than his older brother.

Peploski died in New York, New York in 1972 at age 80. He is buried at Rosedale Cemetery in Linden, New Jersey.
